Now get some idea about the media components needs for PTC. Note: Different plants need different media composition. Their are different types of Media for individual plants. For example, Rooting Media, Shooting media, Callus media, Liquid single cell media and Complete media.

Got fear?? :roll: Commercial base media is also available. I idea about the price, but it can be arranged if you people want. Info: 1 Commercial PTC Unit is located in IMT Manesar. But not allowed visit.

Now, we should move into the main discussion. Those above posts ware to give an Idea about the process and short of introduction of the topic.



Following things we will need to set up a Plant tissue culture facility in our home.

1. An area where we can do Aseptic culture transfer. Preferably a 3x2x2 qft 5 side closed area and with a smooth surface for easy cleaning option. Their must be a good lighting source at the top and an UV lamp for surface and air sterilization. The front should be closed upto 75% with an Acrylic sheet and rest of the open area can be cover with hinge fitted acrylic sheet.

2. Spirit lamp or Bunsen Burner with LPG fittings.

3. Cuttings tools, like forcep, scalpel, needle and scissors

4. Ethanol (70%) for surface cleaning

5. Mercuric chloride solution, 5% Bleaching solution for plant body sterilization

6. Commercial growth Media (MS) and Hormone (Auxin)

7. 5 litter pressure cooker for Sterilization

8. Non absorbent cotton for plugging, containers to keep the plants (Autoclave safe). Preferably polypropylene make.

9. Racks to keep those container after inoculation. Lighting fittings on top with T5 (6500k), Osram fluora

10. Temp 20-25*C in grow-up room

Macronutrients
Ammonium nitrate (NH4NO3) 1,650 mg/l
Calcium chloride (CaCl2 · 2H2O) 440 mg/l
Magnesium sulfate (MgSO4 · 7H2O) 370 mg/l
Potassium phosphate (KH2PO4) 170 mg/l
Potassium nitrate (KNO3) 1,900 mg/l



Micronutrients
Boric acid (H3BO3) 6.2 mg/l
Cobalt chloride (CoCl2 · 6H2O) 0.025 mg/l
Cupric sulfate (CuSO4 · 5H2O) 0.025 mg/l
Ferrous sulfate (FeSO4 · 7H2O) 27.8 mg/l
Manganese sulfate (MnSO4 · 4H2O) 22.3 mg/l
Potassium iodide (KI) 0.83 mg/l
Sodium molybdate (Na2MoO4 · 2H2O) 0.25 mg/l
Zinc sulfate (ZnSO4·7H2O) 8.6 mg/l
Na2EDTA · 2H2O 37.2 mg/l

Little problem in that... I do not have expertness in PTC. Just did some Lab work during my Post Graduation and that is also 8 years back! Hardly remember the media composition. As I mentioned earlier, University note books could help me a lot to even designing a suitable media for Aquatic plants. Considering our aquatic plant a soft tissue one, we can try with any Complete media to start with. According to the response from the plant, we can modify the components we need. Adding Auxin hormone will do the rest.
